摘要
用组织化学定位法检测小白菜 gus基因瞬间表达情况 ,分析三种农杆菌对小白菜的侵染能力 ,结果表明 :AGL 1对小白菜的侵染能力最强 ,EHA10 5次之 ,LBA4 4 0 4最弱 ;通过共培养后定时测定 gus基因瞬间表达情况 ,确定农杆菌与小白菜子叶的最佳共培养时间为 2 3d。
The cotyledons of Brassica chinensis were inoculated with 3 agrobacterium tumefaciens strains(AGL I,EHA105 and LBA4404)in combination with pBin19 derived binary plasmid p35SGUSINT,whitch carrying the NosnptII selection gene.and 35Sgus(intron)report gene.The result showed AGL 1 infected explants had the highist GUS ecpression,while the least GUS activeity were the explants infected with LBA4404.2 or 3days cocultivating were suitable for B.Chinensis cotyledons.
