摘要 目的探讨Aβ40、Aβ42、Aβ42Arc对阿尔茨海默病(AD)转基因果蝇认知功能的影响。方法分别建立Aβ40、Aβ42、Aβ42 Arc转基因AD果蝇模型。根据实验要求将果蝇分为4组,分别是野生果蝇对照组、Aβ40组、Aβ42组、Aβ42 Arc组。在整体动物水平上进行果蝇行为学研究,分别检测各组果蝇的嗅觉短期记忆能力、攀爬能力和平均寿命。结果在巴甫洛夫嗅觉相关短期记忆测试、攀爬能力试验、平均寿命测试中,与对照组相比,3组转基因果蝇的嗅觉记忆能力、攀爬能力、平均寿命均明显降低。结论 Aβ的毒性作用会导致AD转基因果蝇认知功能下降。 Objective To study the effects of β-amyloid proteins Aβ40, Aβ42, and Aβ42Arc on the cognitive functions in the trans- genic drosophila with Alzheimer' s disease (AD). Methods The experiment was designed into four groups : normal wild-type, Aβ40, Aβ42 and Aβ42Arc transgenic drosophila. Pavlovian olfactory memory and climbing abilities and life span were measured in the four groups. Results Compared with the control group, the short-term dfactory memory and climbing abilities as well as life span were reduced in the Aβ40, Aβ42 and Aβ42Arc groups. Conclusions Aβ may result in a decrease in cognitive functions in transgenic dro- sophila with AD.
