摘要
目的观察血管紧张素-(1-7)[Ang-(1-7)]对高糖诱导的小鼠胰岛素分泌细胞株NIT-1氧化应激的影响。方法将对数生长期的NIT-1细胞随机分成A、B、C、D、E组,A组不处理,B、C、D、E组分别用终浓度为10-5mol/L的Ang-(1-7)、35 mmol/L葡萄糖、10-5mol/L的Ang-(1-7)+35 mmol/L葡萄糖、10-5mol/L的Mas受体拮抗剂A-779+10-5mol/L的Ang-(1-7)+35 mmol/L葡萄糖干预72 h。用DCFH-HA荧光染色法检测活性氧簇(ROS),硫代巴比妥酸法测定丙二醛(MDA)。结果 A、B、C、D、E组ROS分别为24.25±1.85、25.31±3.60、33.14±3.79、21.47±1.66、31.71±3.88,MDA分别为(17.99±0.40)、(17.29±1.97)、(30.47±2.61)、(21.57±1.32)、(29.62±3.23)μmol/g;C、E组与A、B、D组比较,P均<0.05;C、E组比较,P均>0.05;A、B、D组比较,P均>0.05。结论Ang-(1-7)通过Mas受体抑制高糖诱导的NIT-1细胞氧化应激。
Objective To observe the effect of angiotensin-(1-7) [ Ang-(1-7) ] on oxidative stress induced by high glucose in mouse insulin-secreting cell line NIT-1. Methods NIT-1 cells in logarithmic phase were divided into five groups: group A with no treatment; the cells in group B, C, D and E were treated with 10^-5 mol/L Ang-( 1-7 ) , 35 mmol/ L glucose, 10^-5 mol/L Ang-(1-7) + 35 mmol/L glucose, receptor inhibitor (A-779, 10^-5 mol/L ) + 10^-5 mol/L Ang-( 1- 7) + 35 mmol/L glucose respectively. Every group was treated for 72 hours. Reactive oxygen species (ROS) was detected by DCFH-DA fluorescent staining assay. Malondialdehyde (MDA) was measured by TBA method. Results Concentrations of ROS in group A, B, C, D and E were 24.25 ± 1.85, 25.31 ±3.60, 33.14 ±3.79, 21.47 ±1.66 and 31.71 ±3. 88 respectively. Concentrations of MDA in group A, B, C, D and E were (17.99 ±0.40) , (17.29 ±1.97), (30.47 ±2.61 ), (21.57 ±1.32) and (29.62 ±3.23 ) μmol/g respectively. Group C and E were statistically different from group A, B and D ( all P 〈 0.05 ). There was no statistical difference between group C and E ( P 〉 0.05 ) , meanwhile, there was no statistical difference among group A, B and D (all P 〉 0.05 ). Conclusion Ang-( 1-7 ) treatment can block the effect of high glucose on oxidative stress in NIT-1 cells via Mas receptor.
