摘要
为研究金针菇漆酶的酶学性质,试验选取金针菇杂交菌株L7号作为试验材料,对其进行非变性聚丙烯酰胺凝胶电泳(Native-PAGE),发现其含有3种漆酶同工酶,同时采用分光光度法,以ABTS[(2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)]为底物测定金针菇杂交菌株漆酶活性的最佳反应条件。结果表明:适宜测定波长为410nm、体系pH4.5、反应温度45℃。
In order to study the lacese enzymatic properties of Flammulina velutipes, L7 as experimental material is hybrid strains of Flammulina velutipes. The Native-PAGE analysis showed that it possessed three kinds of lacease isoenzymes. By using speetrophotometry and choosing ABTS [2,2'-Azino-bis(3-ethy lbenzothiazoline-6-sulfonie acid)I, the activity of lacease can be determined, the results showed that: the appropriate determination of a wavelength 410 nm, system pH 4.5, reaction temperature 45℃.
