水稻OsMED32基因RNA干扰载体的构建

摘要 水稻Mediator complex subunit 32(OsMED32)和哺乳类中的MED29以及酵母中的MED2属于同源基因,是主要的DNA结合转录因子。该研究利用RNAi技术,在克隆得到OsMED32基因全长序列的基础上,选取其中一段保守片段构建RNA干扰表达载体p1300-OsMED32 RNAi,使OsMED32在水稻中表达沉默,并转化至农杆菌EHA105中,为进一步研究OsMED32基因功能奠定了良好的基础。 The OsMED32 and MED29 which in mammalian and the MED2 which in yeast are belonging to homologous genes. The OsMED32 is the major DNA-binding transcription factor.On the basis of the cloned OsMED32 complete sequence, a more conservative fragment was selected to be used for construction of the RNA interference vector p1300-OsMED32 RNAi.As a result, the OsMED32 can not normally expression in rice and conversion to agrobacterium EHA105,which can help us to find out the function of OsMED32.
