肢体缺血预处理对脑缺血大鼠再灌注损伤VEGF、PDGF-B的影响 被引量:5

摘要 目的观察肢体缺血预处理(LIP)对大鼠中动脉缺血再灌注损伤后血管内皮生长因子(VEGF)、血小板源生长因子-B(PDGF-B)的影响,探讨LIP脑保护作用机制。方法健康雄性Wistar大鼠60只,随机分为两组。大鼠中动脉缺血再灌注组(MCAO组):采用线栓法制备MCAO模型;肢体缺血预处理组(LIP+MCAO组):采用改良的无创血压测定仪制备LIP模型后即刻制备MCAO模型。依据缺血后再灌注时间不同,进一步将MCAO组和LIP+MCAO组分为6h、24h、3d、7d、14d五个亚组,每组6只大鼠。对各组大鼠进行神经功能缺损评分后处死。采用HE染色观察大鼠脑组织的病理改变、免疫组织化学法检测脑组织VEGF、PDGF-B、CD31及新生的成熟血管(α-SMA)的表达。结果 LIP可明显减轻MCAO神经功能及组织学损害,与MCAO组比较,LIP+MCAO组VEGF、PDGF-B、CD31及α-SMA的表达明显增加(x±s)。结论 LIP对其后脑缺血再灌注损伤有明显的保护作用,可能与LIP诱导的血管再生有密切关系。
