奥扎格雷联合丹参酮ⅡA对肢体缺血再灌注损伤的保护机制研究 被引量:1

摘要 目的探讨奥扎格雷注射液联合丹参酮ⅡA磺酸钠注射液对肢体缺血再灌注损伤的治疗作用。方法健康成年SD大鼠50只,随机分为模型组、肢体缺血再灌注组(A组)、丹参酮ⅡA组(B组)、奥扎格雷组(C组)、奥扎格雷与丹参酮ⅡA联合组(D组),每组10只,建立大鼠肢体缺血再灌注损伤动物模型,在恢复血流灌注30min前,A组腹腔注射生理盐水,B组腹腔注射丹参酮ⅡA磺酸钠注射液,C组腹腔注射奥扎格雷注射液,D组腹腔注射奥扎格雷及丹参酮Ⅱ磺酸钠A注射液,模型组不给予任何处理。在结扎4h后松解,松解2h后腹主动脉取血,测定血清肌酸激酶(CK)、乳酸脱氢酶(LDH)、肌酸激酶同工酶(CK-MB)、血栓素A2(TXA2)、前列环素(PGI2)的含量,后腿骨骼肌组织及血浆中丙二醛(MDA)、黄嘌呤氧化酶(XOD)、超氧化物歧化酶(SOD)含量。电镜及光镜观察骨骼肌组织结构变化。结果再灌注后联合组LDH、CK-MB、Mb、TXA2、PGI2明显低于其他组(P<0.05),MDA、SOD、XOD差异有统计学意义,光镜及电镜下观察骨骼肌损伤轻于其他组。结论两药联合对缺血再灌注骨骼肌保护作用较好。 Objective To investigate the efficacy of ozagrel injection and sodium tanshinone Ⅱ A sulfonate injection on ischemic reperfusion injury of limbs. Methods Fifty healthy adult SD rats were randomly divided into model group,ischemia reperfusion group treated with saline,tanshinone Ⅱ A group treated with tanshinone ⅡA, ozagrel group treated with ozagrel, and combination group treated with anshinone Ⅱ A and ozagrel (10 rats in each group). Serum dehydrogenase (LDH) ,creatine kinase (CK), Mb, thrombox- ane A2 (TXA2), prostacyclin (PGI2), malonyldialdehyde (MDA), superoxide dismutase (SOD), and xanthine oxidase (XOD) content in skeletal muscle and plasma were tested. The skeletal muscle tissue structure was observed by Optical and electron microscopic. Re- sults LDH,CK, Mb,TXA2 , and PGI2 in reperfusion group were significantly lower than those in other groups(P〈0.05) and MDA, SOD,XOD had remarkable difference. Skeletal muscle injury was lighter than that in other group. Conclusion Ozagrel injection and sodium tanshinone ⅡA sulfonate injection had better protection effect for ischemia reperfusion injury of skeletal muscle.
