重组β-防御素2对大鼠多重耐药性鲍曼不动杆菌肺炎的保护作用

Protective effect of recombinant β-definsin-2 on multiresistant Acinetobacter baumannii pneumonia in rats

摘要 目的探讨大鼠重组β-防御素2(rβD-2)预处理对肺部感染多重耐药鲍曼不动杆菌大鼠的保护作用。方法 64只SD大鼠随机均分为四组:rβD-2重组质粒(A)组、空白载体质粒(B)组、多重耐药鲍曼不动杆菌(C)组和生理盐水对照(D)组。72h后检测血清中白细胞介素1β(IL-1β)、髓过氧化物酶(MPO)和rβD-2水平,RT-PCR检测肺组织中rβD-2mRNA表达,HE染色进行肺损伤病理评估。结果与D组相比,A、B、C组大鼠肺组织rβD-2mRNA的表达量及血清IL-1β、MPO和rβD-2蛋白明显升高(P<0.05)。A组肺组织rβD-2mRNA表达量以及血清IL-1β、MPO蛋白水平较B、C组明显增加(P<0.05)。结论 rβD-2对感染多重耐药鲍曼不动杆菌的大鼠肺组织具有一定程度的保护作用,可能与其杀菌作用及提高IL-1β和MPO的水平有一定关系。 Objeetive To determine if recombinant β-defensin-2(rβD-2) pretreating intranasally can protect the lungs from multiresistant Acinetobacter baumannii infection in rats. Methods Sixty- four SD rats were equally randomized into 4 groups of A(pretreated with pEGFP-NI-r^D-2/PEI), B (with pEGFP-NI-null/PEI), C(with Acinetobacter baumannii)and D (with normal saline). Serum levels of interleukin-ll3(IL-lβ), myeloperoxidase(MPO), rβD-2 were detected at 72 hours later. The expression of rβD-2 mRNA in the lung tissues and histopathological changes of the lungs were measured by RT-PCR and HE staining, respecttively. Results Compared with group D, the expression of rβD-2 mRNA and serum levels of IL-Iβ, MPO, and rβD-2 were all obiviously increased in groups of A,B and C(P〈0. 05). Compared with groups of B and C, serum levels of IL-lfl and MPO and the expression of rβD-2 mRNA were all obiviously increased in group A (P〈0. 05 ). Conclusion rβD-2 can kill multi-resistant Acinetobacter bacillus in vivo and modulate the expressions of IL-1β and MPO. It can protect the lungs against injury induced by multiresistant Acinetobacter baumannii.
