多西环素在兔椎间盘退变中的作用 被引量:1

Effect of doxycycline on intervertebral disc degeneration in rabbits

摘要 目的探讨多西环素在兔椎间盘退变(IVDD)中对基质金属蛋白酶3(MMP-3)表达的影响。方法将20只新西兰大白兔建立IVDD模型后,随机均分为多西环素30mg实验组(A组)和对照组(B组)。术后8、12周,采用MRI检查IVDD情况,免疫组化检测椎间盘组织中MMP-3的表达,HE染色观察椎间盘组织的病理改变。结果与B组相比,A组IVDD程度明显减轻(P<0.05),MMP-3表达降低(P<0.05);B组椎间盘髓核萎缩较A组更为明显,纤维环排列更加混乱。结论MMP-3表达与IVDD程度呈正相关;多西环素能够能抑制MMP-3表达,延缓IVDD进程。 Objective To investigate the inhibitory effect of doxycycline on matrix metalloproteinase-3(MMP-3) expression in the rabbits with intervertebral disc degeneration(IVDD). Methods Twenty New Zealand rabbits with IVDD were equally randomized into two groups of A (injected with doxycycline 30 mg for 4 weeks) and B(injected with equal normal saline for 4 weeks). At 8 and 12 weeks after operation, IVDD status was observed by MRI examination, and MMP-3 expression in the intervertebral disc tissues was detected by immunohistochemistry. The pathological changes of intervertebral disc tissues were evaluated by HE staining. Results Compared with group B, the extent of IVDD was significantly improved (P〈0. 05), MMP-3 expression was decreased (P〈0. 05) in group A. Nucleus pulposus atrophy in intervertebral disc was more serious and anulus fibrosus array was more disordered in group g than those in group A. Conclusion MMP-3 expression is positively correlated with the extent of IVDD. Doxyeycline may delay IVDD progression via inhibiting MMP-3 expression.
