摘要
目的探讨亚低温对脑出血后内源性神经干细胞(neural stem cells,NSCs)增殖及迁移的影响。方法采用胶原酶脑内立体定向注射制作脑出血模型。模型成功后将大鼠随机分为对照组和亚低温组,每组分为7、14、21、28 d亚组。应用腹腔注射溴脱氧尿嘧啶核苷(Bromodeoxyuridine,BrdU)标记增殖的NSCs。采用免疫组织化学方法动态观察同侧侧脑室下区(subventricular zone,SVZ)及血肿周围BrdU阳性细胞的变化。结果对照组同侧SVZ及血肿周围BrdU阳性细胞在出血后7 d时开始增加,14 d达到高峰,21 d后开始下降。亚低温组显著增加各时间点SVZ及血肿周围BrdU阳性细胞(P<0.01)。结论亚低温能促进脑出血后内源性NSCs增殖及迁移。
Objective To investigate the influence of mild hypotherrnia on the proliferation and migration of neural stem cells (NSCs) in rats with intraeerebral hemorrhage(ICH). Methods ICH was induced by an injection of collagenase into the striatum of male Sprague-Dawley rats. Animals were randomly divided into control group and mild hypothermia group. Each group was divided into 7 d, 14 d, 21 d, 28 d subgroups respectively. Bromodeoxyuri- dine (BrdU) was injected intraperitoneally to label the proliferative NSCs. Immunohistochemical staining was used to observe the BrdU positive cells in SVZ and peri-hemorrhage area. Results In control group, the number of BrdU-pos- itive cells in the ipsilateral SVZ and peri-hemorrhage area increased at 7day, peaked at 14 day, decreased at 21 day. Compared with control group, mild hypothermia treatment increased the number of BrdU-positive cells in SVZ and peri-hemorrhage area at every time point. Conclusions Mild hypothermia can promote the proliferation and migration of endougenous NSCs after intraecerebral hemorrhage.
