高效液相色谱法测定婴幼儿乳制品中免疫球蛋白IgG的含量

本文建立了一种测定婴幼儿乳制品中免疫球蛋白IgG含量的方法。针对前处理方法及高效液相色谱条件进行实验研究,确定了提取溶剂、离心速率、超声时间、样品稀释倍数、调pH值所用酸及检测波长、色谱柱、流速、柱温、流动相的pH值的参数值,采用保留时间定性,峰面积外标法定量。方法检出限为0.7μg.mL-1,回收率为92.16%,标准偏差3.34%,IgG浓度在0.1~1.0mg.mL-1范围内与其色谱峰面积之间为线性关系：y=2519x-24.71（R2=0.9992）。对实际样品中的IgG含量进行了测定。本文方法所获得的色谱峰分离度优于国标方法。
