摘要
目的探讨新疆芍药中总多糖的最佳提取工艺,初步研究新疆赤芍总多糖对变异链球菌的作用。方法采用蒽酮比色法,通过水浸提、乙醇沉淀总多糖、Sevage脱蛋白、干燥等方法,应用正交试验设计,测定总多糖的含量。应用连续稀释法和琼脂扩散法研究芍药总多糖对变异链球菌的作用。结果新疆芍药总多糖最佳提取工艺为料液比1∶8、浸提次数为2次、浸提时间1.5h,得总多糖含量为51.57%;新疆芍药总多糖对变异链球菌的抑制作用:MIC为110.4mg.mL-1。结论新疆芍药总多糖对变异链球菌生长具有一定的抑制作用,可作为口腔疾病治疗的参考中药。
Objective To explore the best extraction conditions of polysaccharides from Paeonia sinjiangertsis K. Y. Pan., preliminary study the effect of polysaecharides on Streptococcus mutans. Methods The separation procedure of precipitation with alcohol and the purification from the removing proteins were deeply analyzed. Meanwhile the contents of polysaccharide were determined by Anthrone colorimetry through an orthogonal experiment (L9(3^4) ). Determined antibacterial activities of polysaceharides from Paeonia sinjiangensis K. Y. Pan. by serial dilution method and agar diffusion method ( modified Kirby - Bauer method). Results The highest yield was obtained when ratio of solvent to raw material, extraction times, extraction time were 8 : 1, 2, 1.5 h, respectively. The content of soluble polysaceharide is 51.57%. Conclusions The polysaccharides from Paeonia sinjiangensis K. Y. Pan. have certain antibacterial activity for Streptococcus mutans growth.
