温肾方对亚临床甲状腺功能减退模型大鼠心肌组织T3及甲状腺激素受体的影响 被引量:8

Effect of Wenshen Prescription on Myocardial Tissue T3 and Thyroid Hormone Receptors of Subclinical Hypothyroidism Model Rats

摘要 目的:探讨温肾方对亚临床甲状腺功能减退(甲减)模型大鼠心肌组织T3及甲状腺激素受体的影响。方法:采用"甲状腺全切术+术后皮下注射L-T4"法制备亚临床甲减大鼠模型,设定空白对照组、模型组、中药低剂量组、西药组、中西药组及中药高剂量组,用ELISA法参照组织T3试剂盒说明测定心肌组织T3水平,RT-PCR法测定心肌组织甲状腺激素受体表达情况,同时观察温肾方对其指标的干预情况。结果:温肾方能提高心肌组织T3水平,尤以中药高剂量组最为显著,中药高剂量组优于单纯中药低剂量组、西药组及中西药组,接近空白对照组;温肾方在下调Tα2表达,上调Tβ1表达方面有效(P<0.05);中西药组在Tα1表达调节上优于单纯中药组。结论:亚临床甲减存在的心肌组织内甲减,可能是导致心脏损害的机制之一;温肾方能显著提高心肌组织内T3水平,作用优于L-T4,能改善心肌组织内甲状腺激素受体基因表达,从而具有防治亚临床甲减心脏并发症的作用,为中药治疗亚临床甲减防治并发症提供实验依据,温阳补虚法可能为有效治疗亚临床甲减的方法之一。 objective:to study the subclinical hypothyroidism on subclinical hypothyroidism(JiaJian) model rats myocardial tissue and thyroid hormone T3 receptors.Methods:"full thyroid postoperative subcutaneous injection technique+L-T4" preparation of subclinical JiaJian rats model,set up blank control group,model group,low dose group,western medicine group,the Chinese and western medicine group and high dose group and reference group T3 with ELISA method kit T3 levels in myocardial tissue,rt-pcr method expression in myocardial tissue of thyroid hormone receptors,while the index of the intervention on the observation of the kidney and party.Results:the kidney can improve the level of myocardial tissue T3,especially in high dose group of the most significant Chinese medicine,Chinese medicine high dose group is superior to the pure Chinese medicine,western medicine group and low dose group of+western medicine group,close to the blank control group;Kidney T alpha 2 expression in lower,raising T expressed in beta 1 effective(P0.05);T+western medicine group in alpha 1 on expression regulation is better than that of pure Chinese medicine group.Conclusion:subclinical JiaJian JiaJian in myocardial tissue,one is likely to cause heart damage mechanism;The kidney can significantly improve the level of myocardial tissue in T3,effect is better than that of L-T4,can improve the myocardial tissues of thyroid hormone receptor gene expression,which have the function of the prevention and treatment of subclinical JiaJian cardiac complications,for traditional Chinese medicine treatment of subclinical JiaJian prevention and treatment of complications to provide experimental basis,explored tonify deficiency may have as one of the effective treatment of subclinical JiaJian method.
