异丙肾上腺素致心肌纤维化大鼠心肌MCP-1和ICAM-1的表达 被引量:1

摘要 目的研究异丙肾上腺素(1SO)诱导大鼠心肌纤维化后单核细胞趋化蛋白-1(MCP—1)、细胞间黏附分子-1(ICAM-1)等表达的变化,探讨炎性因子在心肌纤维化发生发展中的作用。方法16只雄性sD大鼠随机分为对照组、模型组,每组8只。模型组皮下注射ISO5mg·k^-1·d^-1×7天,对照组皮下注射等量生理盐水7天,于实验第5周末处死动物留取心脏标本。实验末各组取心脏计算心重指数(heartweightindex,HWI),左心室心肌组织行Masson染色测胶原容积积分(CVF);ELISA法检测心肌组织MCP-1和ICAM-1的水平;RT—PCR测定心肌组织MCP-1mRNA和ICAM-1mRNA表达。结果与对照组相比,模型组大鼠HWI及左心室心肌组织CVF明显升高(P〈0。01);模型组心肌组织中MCP-1mRNA和蛋白及ICAM-1mRNA和蛋白的表达均明显升高(P〈0.01)。结论炎性因子MCP-1和ICAM-1的过度表达参与了ISO诱导大鼠心肌纤维化的发生发展。
