摘要
SIRT1是一种二氢尿嘧啶脱氢酶依赖的具有去乙酰化酶活性的多功能转录调节因子。为分析体外白黎芦醇和尼克酰胺诱导的猪卵巢颗粒细胞中SIRT1 mRNA表达量的变化与颗粒细胞分泌雌激素的相关性,采用qRT-PCR方法进行SIRT1 mRNA分析,猪雌激素酶联免疫分析试剂盒检测培养液中雌激素浓度。结果表明,培养液中白黎芦醇浓度达到50μmol/L时,颗粒细胞中SIRT1 mRNA表达量极显著高于空白对照组(P<0.01);相反,尼克酰胺浓度达到50μmol/L时,处理组颗粒细胞中SIRT1 mRNA表达量极显著低于空白对照组(P<0.01);培养液中,颗粒细胞分泌雌激素的变化规律与SIRT1 mRNA表达规律一致,相关分析表明,两者存在较强的正相关,相关系数为0.966 8。
Sirtuin 1 (SIRT1), the mammalian homolog of Sir2, is a dihydrouracil dehydrogenase-dependent deacetylase, which plays an important role in a variety of biological processes, including cell proliferation and differentiation, apoptosis, and metabolism. In this study, correlation between estrogen secretion and SIRT1 gene expression level in- duced by resveratrol and nicotinamide was studied. SIRT1 expression level was analyzed by qRT-PCR and estrogen secretion level was tested by enzyme-linked immunosorbent assay kit. The results showed that when resveratrol concentration reached 50 μmol/L in cuhure liquid, expression level of SIRTI mRNA was significantly higher than that in control (P〈0. 01 ). On the contrary, when nicotinamide concentration reached 50 μmol/L, the level was significantly lower than that in control (P〈0. 0! ). Meanwhile, the change rules of estrogen secretion level were similar to those of SIRT1 mRNA expression level in granulosa cells. Correlation analysis revealed that there was a strong positive correlation be- tween estrogen secretion and SIRT1 gene expression level, with the correlation coefficient being 0. 966 8.
