HPLC法测定大蓟配方颗粒中蒙花苷和柳穿鱼叶苷的含量

HPLC method for determining content of linarin and pectolinarin in Cirsium japonicum formula granules

目的:建立大蓟配方颗粒中蒙花苷和柳穿鱼叶苷的含量测定方法。方法:采用HPLC法,Hypersil ODS2 C18色谱柱(250mm×4.6mm,5μm);乙腈-0.1%磷酸(23∶77)为流动相;体积流量1mL/min;检测波长330nm;柱温30℃。结果:蒙花苷对照品在0.044～0.44μg进样量与峰面积呈良好的线性关系(r=0.9997),平均加样回收率为100.26%,RSD为1.25%(n=6);柳穿鱼叶苷对照品在0.141～2.115μg进样量与峰面积呈良好的线性关系(r=0.9999),平均加样回收率为100.37%,RSD为1.18%(n=6)。结论:该方法操作简便,结果准确,重现性好,可用于大蓟配方颗粒的质量控制。

Objective：To establish a content determination method for linarin and pectolinarin in Cirsium japonicum formula granules.Methods：High-performance liquid chromatography（HPLC） was performed on a C18 column（Hypersil ODS2,250 mm × 4.6 mm,5 μm） with a mobile phase of acetonitrile-0.1% phosphoric acid（23：77） at a flow rate of 1 ml/min,a detection wavelength of 330 nm,and a column temperature of 30℃.Results：The reference substance for linarin showed a good linear relationship between amount and peak area in the range of 0.044-0.44 μg（r = 0.9997）;the average recovery rate was 100.26%,and the relative standard deviation（RSD） was 1.25%（n =6）.The reference substance for pectolinarin showed a good linear relationship between amount and peak area in the range of 0.141 2.115 μg（r = 0.9999）;the average recovery rate was 100.37%,and the RSD was 1.18%（n = 6）.Conclusion：The HPLC method is easy to operate,accurate,and highly repeatable and can be used for the quality control of Cirsium japonicum formula granules.
