摘要
目的观察氨基甾体H51817抑制白血病K562细胞系增殖和诱导其分化的作用并探讨其作用机制。方法用细胞计数、MTT、集落培养的方法观察氨基甾体H51817抑制K562细胞增殖的能力;用联苯胺染色、Wright染色、流式细胞仪检测氨基甾体H51817诱导K562细胞分化的作用;用原子吸收光谱仪检测细胞内外钙离子浓度([Ca2+])的变化。选取阿糖胞苷(cytosine arabinoside,Ara-C)和吲哚美辛(Indomethacin,IN)作为阳性对照。结果浓度为10-5mol/L的氨基甾体H51817可以明显的抑制K562细胞的生长(P<0.01),并且可以诱导其向红系分化,同时使K562细胞内[Ca2+]降低(P<0.05)。结论氨基甾体H51817能有效地抑制K562细胞的增殖和诱导其向红系分化,其作用机制可能与K562细胞内[Ca2+]降低有关。
Objective To explore the proliferation and differentiation inducing effects and potential mechanisms of aminosteroid H51817 on leukemia K562 cell line.Methods Proliferation was detected by cell counting,MTT assay and semisolid cultures.Differentiation was detected by morphology,benzidine staining and flow cytometry.The changes of [Ca2 +]were measured by atomic absorption spectrometer.Took cytosine arabinoside and indomethacin as the positive controls.Results Aminosteroid H51817 inhibited proliferation of K562 cells respectively at 10-5mol/L(P 0.01),induced K562 cells toward erythroid differentiation,and decreased intracellular [Ca2 +]in K562 cells(P 0.05).Conclusion Aminosteroid H51817 could inhibit the proliferation of K562 cells and induce them toward erythroid differentiation.The effects of aminosteroid H51817 on K562 cells maybe associated with the decrease of intracellular [Ca2 +]in K562 cells.
