下瘀血汤组分配伍抗免疫性肝纤维化大鼠模型的实验研究

Experimental study of Xiayuxue Decoction component compatibility aganist immunity hepatic fibrosis rat model

目的观察下瘀血汤组分配伍抗免疫性肝纤维化大鼠模型的作用,为组分配伍的研究提供实验依据。方法 Wistar大鼠随机分为正常组、造模组,造模组大鼠腹腔注射猪血清复制肝纤维化模型,每次0.5 ml/只,每周2次,共12周。造模成功后造模组大鼠随机分为模型组、下瘀血汤组分配伍组,第8周末开始下瘀血汤组分配伍组大鼠造模同时灌胃给药,按10 ml.kg-1容积,每日0.648 g.kg-1,连续4周。正常组、模型组大鼠给予同体积生理盐水。12周末,处死动物,获取标本,测定血清肝功能、肝组织羟脯氨酸(Hyp)含量;realtime-PCR检测肝组织TGF-β1mRNA表达。结果与模型组比较,下瘀血汤组分配伍组大鼠血清总蛋白(TP)、球蛋白(Glb)含量明显降低(P<0.05),白蛋白(Alb)含量升高,A/G比值明显改善(P<0.05);与模型组相比,下瘀血汤组分配伍组TGF-β1mRNA的表达明显降低(P<0.05)。结论下瘀血汤组分配伍对大鼠免疫性肝纤维化模型具有保护肝细胞功能,其机制可能是通过抑制TGF-β1mRNA表达而发挥抗肝纤维化作用。

Objective To observe Xiayuxue decoctiong ingredients formula against immunity liver fibrosis rats model,for ingredients recipe research provide experimental basis. Methods Wistar rats were randomly divided into normal group, model group and rat fiver fibrosis model was duplicated intraperitoneal injection of porcine serum. After successful modeling, rats were randomly divided into model group, Xiayuxue decoction group and its ingredients recipe group. Rats were given TCM preparation for 4 weeks, at the end of 12 weeks, animals were sacrificed to obtain liver tissue samples for determination of Hyp content and TGF - β1 mRNA mRNA expression. Results Compared with model group, TP and Glb content significantly （ P 〈 0. 05 ） decreased in Xiayuxue decoction＇s ingredients recipe group, A/G ratio increased significantly （P 〈 0.05 ） ; Alb content increased, but the difference was not statistically significant （P 〉 0.05 ）. And compared with model group, TGF- β1mRNA mRNA expression in ingredients recipe group decreased significantly （P 〈 0.05 ）. Conclusion Xiayuxue decoction＇s ingredients recipe protective liver cell function against rat immunity liver fibrosis model, the mechanism may be by inhibiting TGF - β1 mRNA expression to play a role of liver fibrosis resistance.
