摘要
目的通过观察不同针灸预刺激对阿尔茨海默病(AD)模型大鼠热休克蛋白HSP70、HSP90的影响,以探讨针灸防治AD的作用机理。方法将SD大鼠随机分为6组,即正常组、假手术组、模型组、预艾灸组、预电针组、预针灸组,每组各lO只,以双侧海马注入Aβ1-42制作AD大鼠造模。取穴"百会"和双侧"肾俞"穴。采用免疫组化法测定大鼠海马CA1区HSP70、HSP90的平均光密度表达。结果通过4周的预艾灸治疗,预艾灸组、预电针组、预针灸组大鼠海马CA1区HSP70、HSP90的平均光密度表达均较模型组有显著提高(P<0.01)。结论艾灸预刺激、电针预刺激及针灸预刺激能通过诱导脑内HSP70、HSP90的表达来抑制Aβ毒性级联反应,从而起到保护神经元的作用。
Objective By investigating the effect of difference pre-stimulation with acupuncture-moxibustion on heat shock proteins in rat models of Alzheimer's disease,so as to research into its underlying mechanism in preventing and controlling AD in the rats.Methods All animals were randomly divided into,normal group,sham-operation group,model group,pre-moxibustion group,pre-acupuncture group and pre-acupuncture and moxibustion group.AD rats were induced by injecting Aβ1-42 into the double hippocampus.We performed pre-stimulation methods on two acupoints,Baihui(DU20) and Shenshu(BL23).Meanwhile,immunohistochemistry was used to determine the levels of expression of rat's HSP70 and HSP90 in rat's hippocampus area.Results Compared with model group,all pre-stimulation groups rats increased obviously in Hsp70 and Hsp90 expression of hippocampal area(P 0.01).Conclusion These results imply that pre-stimulation with acupuncture and moxibustion could induce the expression of heat shock proteins in hippocampus area,inhibit consequently the Aβ Toxic cascade,to protect the function of neurons in pathological changes of Alzheimer's Disease.
