九味白术汤对溃疡性结肠炎大鼠血清TNF-α及IL-4水平的影响

Effect of Jiuwei Baizhu Decoction on Serum Levels of TNF-α and IL-4 in Ulcerative Colitis Rats

目的:观察九味白术汤(JWBZ)对溃疡性结肠炎(UC)大鼠血清TNF-α及IL-4水平的影响。方法:48只大鼠完全随机分为正常对照组,模型组,柳氮磺胺吡啶(SASP)组,九味白术汤高、中、低剂量组;采用2,4,6-三硝基苯磺酸(TNBS)复制大鼠溃疡性结肠炎;连续给药10 d后,观察UC大鼠结肠黏膜损伤指数(CMDI)、血清TNF-α及IL-4水平的变化。结果:CMDI评分:模型组大鼠显著高于正常对照组(P<0.01),JWBZ低、中、高剂量组及SASP组均显著低于模型组(P<0.05或P<0.01),JWBZ中剂量组显著低于SASP组(P<0.05);血清TNF-α水平:模型组显著高于正常对照组(P<0.01),JWBZ低、中、高剂量组及SASP组显著低于模型组(P<0.01),JWBZ中剂量组显著低于SASP组;血清IL-4水平:模型组显著低于正常对照组(P<0.01),JWBZ低、中、高剂量组及SASP组显著高于模型组(P<0.01),中剂量组显著高于SASP组(P<0.01),而低剂量组显著低于SASP组(P<0.05)。结论:JWBZ对UC大鼠结肠损伤具有良好的修复作用,其机制可能通过调节促炎因子TNF-α与抗炎因子IL-4的平衡实现。

A Objective ： To observe the effect of Jiuwei Baizhu Decoction （ JWBZ ） on serum levels of interleukin-1β （TNF-eL ）and interleukin-10 （IL-4）in ulcerative colitis rats. Methods：d8 rats were randomly divided into 6 groups ： normal group, model group, salazosulfapyridine （ SASP ） group, low-, middle- and high-dose JWBZ groups. Ulcerative colitis rat model was copied by 2,4,6-TNBS.After 10 days＇ treatment, the changes of colonic mueosal damage index （ CMDI ）, serum levels of TNF- α and IL-4 were observed. Results ： Compared to normal group, the CMDI of model group was significantly increased （ P〈0.01 ）; compared to model group, the CMDI of treatment groups was decreased significantly （ P〈0.05 or P〈0.01 ）; compared to SASP group, the CMDI of middle-dose JWBZ group was significantly decreased （ P〈0.05 ） .Compared to normal group, the serum TNF- α level of model group was increased significantly （ P〈0.01 ）, while IL-4 level was decreased significantly （ P〈0.01 ） .The serum TNF- α level of treatment groups was decreased significantly （ P〈0.01 ）, compared with model group, while IL-4 level was increased significantly （ P〈0.01 ） .Compared to SASP group, the serum TNF- α level of middle-dose JWBZ group was decreased significantly （ P〈0.01 ）; the serum IL-4 level of middle-dose JWBZ group was increased significantly （ P〈0.01 ）, while it decreased significantly of low-dose JWBZ group （ P〈0.05 ）. Conclusion ： JWBZ treated UC rats effectively by regulating the balance of pro-and anti-inflammatory cytokines.
