云芝糖肽对环磷酰胺所致免疫抑制小鼠免疫保护作用 被引量:2

Protective Effects of PSP on Immunosuppression Mice Induced by Cyclophosphamide

摘要 目的:观察云芝糖肽(polysaccharopeptide,PSP)对环磷酰胺(Cyclophosphamide,Cy)所致免疫低下小鼠的免疫保护作用。方法:32只雄性Balb/C小鼠随机分为4组,每组8只。PSP低、高剂量组分别予以相应浓度的PSP溶液灌胃,正常对照组及模型组予以生理盐水灌胃作为对照,连续25 d。用药第17天和第21天除正常对照组外分别予以腹腔注射环磷酰胺制备小鼠免疫低下模型,观察小鼠外周血WBC、淋巴细胞亚群(CD3+CD4+、CD3+CD8+、CD3-CD19+)及胸腺和脾指数变化。结果:用药第22天PSP低剂量组白细胞下降幅度较小,与模型组相比有统计学差异;PSP低、高剂量组的淋巴细胞亚群数目下降水平与模型组相比也明显减少,差别有统计学意义。第26天PSP低、高剂量组WBC均较模型组明显改善,与正常对照组相比无统计学差异;PSP低剂量组淋巴亚群细胞数目显著增加与模型组相比有统计学差异;模型组CD3+CD4+淋巴细胞与PSP高剂量及正常对照组相比均无统计学差异;各组小鼠脾脏及胸腺指数无统计学差异。结论:PSP体内发挥免疫调节作用的机制可能是通过抑制小鼠WBC减少、调节T淋巴细胞亚群、增加B淋巴细胞数量。 Objective : To investigate protective effects of PSP on immunosnppression mice. Methods : 32 Balb/c mice were randomly divided into 4 groups. The mice in normal control group and model group were administrated with saline water, the high dose PSP group ( 500 mg/kg ) and the low dose PSP group ( 125 mg/kg )were received PSP treatment for 25 days, once a day. On the 17th day and 21st day, all groups except normal control group were injected with cyclophosphamide ( 150 mg/kg, i.p. ) to induce the model. The body weight, thymus weigh and spleen weight were analyzed. The absolute numbers of CD3+ CD4+T cells, CD3+CD8+T cells, CD19+ cells and the numbers of WBC in peripheral blood were detected by flow cytometer and haemacytometer. Results : On the 22nd day, compared with the model group, the low dose PSP group improved the numbers of WBC.PSP-treated groups increased the absolute numbers of CD3+ CD4+T cells, CD3+CD8+T cells, CD19~ cells in peripheral blood compared with the model group. The 26th day, PSP-treated mice restored the absolute number of WBC. The low dose of PSP significantly improved the total numbers of T-lymphocyte subsets and B-lymphocytes .The absolute numbers of CD3+CD4+T cells in the model group restored to the normal level. PSP had no significant effect on the weight and indexes of thymus and spleen. Conclusion : The immunoregulatory mechanism of PSP is related to restoring the number of WBC, regulating the total numbers of T-lymphocyte subsets, and improving the numbers of B-lymphocytes.
