摘要
目的:研究运动对RBP4诱导的胰岛素抵抗鼠肝脏PTP1B、PEPCK表达的影响,揭示运动改善RBP4诱导的胰岛素抵抗肝脏机制。方法:雄性SD大鼠给予重组RBP4腹腔注射建立胰岛素抵抗鼠模型,设RBP4+运动组(RE)、RBP4+安静组(RC)和正常安静对照组(C),RE组进行8周游泳训练。ELISA、放射免疫法检测血清RBP4、胰岛素,评估胰岛素敏感指数;免疫组化检测肝脏PTP1B、PEPCK表达,测定肝糖原含量、PEPCK活性。结果:RBP4注射组血清RBP4显著高于C组,胰岛素敏感指数显著低于C组;RE组血清RBP4显著低于RC组,胰岛素敏感指数显著高于RC组,肝脏PTP1表达、BPEPCK表达和活性显著低于RC组,大鼠肝糖原含量显著高于RC组。结论:运动能够纠正RBP4诱导的肝脏PTP1B表达、PEPCK表达和PEPCK活性增加,增加肝糖原含量,抑制内源性葡萄糖的产生,改善胰岛素抵抗。
Objective: To study effects of swimming on PTP1B,PEPCK expression and activity of liver in RBP4-induced insulin resistance rats and explore the liver mechanism that exercise influenced RBP4-induced insulin resistance.Methods: RBP4-induced insulin resistance SD male rats model established by intraperitoneal injection with Recombination RBP4.Model rats divided in RBP4+swimming group(RE)and RBP4+quiet group(RC).Normal control group(C)established too.ELISA,RIA detected serum RBP4,insulin and assessed insulin sensitive index(ISI).IHC detected PTP1B,PEPCK expression in liver.Hepatic glycogen and PEPCK activity were detected in liver.Results: Serum RBP4 of rats increased more in RBP4 intraperitoneal injection exer cise groups than group C.ISI of rats decreased more in RBP4 intraperitoneal injection exercise groups than group C.Serum RBP4 of rats decreased more in group RE than group RC.ISI of rats increased more in group RE than group RC.PTP1B expression,PEPCK expression and activity of liver decreased more in group RE than group RC.Hepatic glycogen increased more in group RE than group RC.Conclusion: Exercise could correct RBP4-induced PTP1B ex pression,PEPCK expression and activation,increase hepatic glycogen and refrain from endogenous glucose production,improve insulin resistance.
