摘要
用基因工程重组技术获得的丙肝病毒 (HCV)核心蛋白抗原与鼠血清白蛋白交联后免疫Balb/c小鼠 ,用杂交瘤技术成功地建立了 4株稳定分泌抗核心抗原单克隆抗体的杂交瘤细胞。试验结果表明 ,该 4株McAbs与免疫抗原及核心区C33肽、CP9、CP1O抗原有较强的抗原 抗体反应 ,与HCVNS3、NS4、NS5无反应 ,在竞争ELISA中 ,对HCV IgG阳性血清有较好的抑制作用。 4株McAbs中 3株为IgG 2a亚类 。
Bal b/c mice were immunized with recombinant HCV core antigen and coupled with murine serum albumin. Four monoclonal antibodies (1F5, 2C5, 1D10, 2G2) with practical value were obtained by hybridoma technique. The reactions of the 4 McAbs with the recombinant core antigen and core antigen C33 peptide, CP9, CP10 were strong, but no reactions were detectable with HCV NS3, NS4, NS5. The 4 McAbs inhibited anti-HCV-IgG positive sera in competitive ELISA. The results also showed that 3 of the 4 monoclonal antibodies were IgG2a and one was IgM in nature.
