摘要 以MTS染色法测定实验剂量的Lipofectin对细胞的毒性作用。以 β 半乳糖苷酶基因为报告基因 ,通过Lipofectin而转染 ,用X gal染色法 ,测定转染效率。结果表明实验剂量 (10 μg/ml)的Lipofectin对细胞生长无明显毒性。Lipofectin对多数肾细胞癌细胞的转染是有效的 ,且转染效率随Lipofectin浓度的增高 (2 .5~ 10 μg/ml)而增高。说明Lipofectin可安全。 To evaluate the efficiency of cationic liposome on human renal cell carcinoma (RCC) cell line. The cytotoxicity of Lipofectin in vitro was determed by colorimetric method. The transfectin efficiency was assessed by beta-galactosidase reporter gene expression and X-gal staining. No significant cytotoxicity of Lipofectin to cultured cell was observed. Lipofectin showed dose-dependent efficient transfection in wide variety of RCC cell lines.The results show that safe and effective gene transfer can be achieved using cationic liposome.
