右美托咪定鞘内注射对大鼠急性疼痛行为及脊髓GIRK2表达的影响 被引量:6

摘要 目的观察鞘内注射右美托咪定(Dex)对切口痛模型大鼠诱发疼痛及脊髓背角GIRK2表达的影响。方法采用大鼠切口痛模型。健康成年雄性SD大鼠60只,随机分为对照组(I组,不作任何处理)、手术组(S组,行左足底切开手术)、安慰剂组(P组,先鞘内注射生理盐水后行左足切开手术)、实验1组(L组,先鞘内注射Dex 0.75μg/kg后行左足切开手术)和实验2组(H组,先鞘内注射Dex 1.5μg/kg后行左足切开手术)。使用vonfrey细丝法观察不同时间点大鼠机械痛阈的改变,免疫荧光技术检测脊髓背角GIRK2蛋白表达的变化。结果L、H组机械痛阈值较S组及P组明显升高(P<0.05);大鼠脊髓背角GIRK2蛋白表达也明显增多(P<0.01)。L组与H组比较差异无统计学意义(P>0.05)。结论鞘内预先给予Dex能减轻大鼠急性创伤后的疼痛阈值,增加大鼠脊髓内GIRK2表达,提示DEX的镇痛作用可能与通过上调GIRK2蛋白表达有关。
