摘要 目的建立同时测定香附、苍术挥发油中α-香附酮和苍术素含量的高效液相色谱法。方法以Diamonsil C18柱(150 mm×4.6 mm,5μm)为色谱柱,以甲醇-水(74∶26)为流动相,α-香附酮和苍术素的检测波长分别为252 nm和340 nm,流速为1.0 mL/min,柱温为25℃。结果质量浓度线性范围α-香附酮为0.25~15.96μg/mL(r=0.999 9),苍术素为1.55~99.00μg/mL(r=0.999 9)。结论该方法准确、简便,可用于香附、苍术混合挥发油中α-香附酮和苍术素含量的同时测定。 Objective To establish a HPLC method for the simultaneous determination of α- cyperone and atractylodin contents in the volatile oil extracted from Rhizoma Cyperi and Rhizoma Atractylodis. Methods The Diamonsil C18 column(150 mm ×4. 6 mm,5 μm) was used as the analytical column. The mobile phase was methanol-water(74:26). The detection wavelength of α-cyperone and atractylodin was at 252 nm and 340 nm,respectively. The flow rate was 1.0 mL/min and the column temperature was at 25 ℃. Results The linear calibration curves of α - cyperone and atractylodin were obtained in the concentration range of 0. 25 - 15.96 μg/mL (r=0.999 9)and 1.55-99.00 μg/mL(r=0.999 9),respectively. Conclusion The established HPLC method is simple,accurate,and can be used to simultaneously determine the content of α- cypemne and atractylodin in the mixed volatile oil extracted from Rhizoma Cyperi and Rhizoma Atractylodis.
