基于SMMC-7721肝癌细胞生长抑制的两种败酱草药效比较及提取方法研究 被引量:11

摘要 目的中药败酱草临床常出现在治疗肿瘤的复方中,但来源较杂,药效难以保证,明确对肝癌细胞生长抑制较好的败酱草品种并优化其提取工艺。方法采用以肝癌细胞抑制率及总黄酮含量为评价指标,筛选两种不同来源的败酱草,正交试验法确定提取工艺,对结果进行分析,结合SPSS17.0软件对考察指标进行相关性分析。结果白花败酱体外抗肿瘤抑制作用高于黄花败酱;白花败酱以SMMC-7721细胞抑制率为指标的提取工艺与以总黄酮含量为指标的提取工艺结果一致均为:10倍量70%乙醇回流提取3次,每次2h,且两种指标相关性良好。结论通过体外药效学比较,白花败酱抑制SMMC-7721肝癌细胞生长优于黄花败酱,中药白花败酱草提取工艺科学合理,为合理利用败酱草奠定了基础。 Objective To choose the sort of patrinia having better inhibition rate of hepatocarcinoma cells and optimize it's extraction process due to the sorts of patriniaits that commonly used in Chinese herbal compound prescription for tumor are complicated and the medicine effect is difficult to guarantee. Methods Two kinds of different sorts of patrinia were selected by taking inhibition rate of human SMMC-7721 hepatocareinoma cells and total flavonoids as indexes,and the extraction process was optimized by using orthogonal test. The correlation of the two indexes was analyzed by SPSS 17.0 software. Results The inhibition rate of Patrinia villosa was better than that of Patrinia scabiosaefolia Fisch. The consistent extracting condition was gotten by using SMMC-7721 human hepatocarcinoma cells as index and total flavonoids as index. And the best extract condition was as follows:ten times the amount 70% ethanol reflux extraction three times,each time 2 h. There was a good correlation between the two indexes. Conclusion The inhibition rate of Patrinia villosa against SMMC-7721 hepatocarcinoma ceils is better than that of Patrinia scabiosaefolia Fisch through contrast on extrinsic medicine effect. The extraction process of Patrinia villosa is scientific and reasonable,which laies a foundation for its clinical use.
