摘要
用Operon公司OPA - 0 1~OPJ- 2 0 2 0 0个 1 0碱基对随机序列引物对甘蓝型油菜隐性核不育涪优AB ,显性核不育 37AB ,广恢系 98- 80 2 6 ,98- 3749及F1材料基因组DNA进行RAPD扩增的结果 ,共获得 1 334条扩增带 ,平均每引物扩增带约 6 .7条 ,表明油菜存在较为丰富的遗传多样性。引物OPA - 0 5 ,OPA - 0 9,OPA - 1 2 ,OPA - 1 3,OPA - 2 0等分别扩增出了大小不同的多态性带 ,初步认为OPA - 1 2 950 是 37A ,OPA - 2 0 750 是涪优A的不育基因分子标记 ,OPA - 0 5 150 0 ,OPA - 1 33 550 是与育性基因有关的带 ,OPA - 1 32 3 0 0 是甘蓝型油菜材料的一条特征带。
Two hundred random 10 bp-primers from Operon company were used in RAPD amplification of genome DNAs of the dominant genetic male sterile line 37AB, the recessive genetic male sterile line Fuyou AB, the poly-restorer lines 98-8206 and 98-3749 and their F 1s. A total of 1334 amplified bands were obtained, averaging 6.7 bands per primer, suggesting that abundant genetic plymorphisms exist in Brassica napus L. Primers OPA-05, OPA-09, OPA-12, OPA-13 and OPA-20 amplified several bands of different milecular weight. OPA-12 950 was the marker for the dominant genetic male sterile gene of 37A, and OPA-20 750 the marker for the recessive genetic male sterile gene of Fuyou A. OPA-05 1500 and OPA-13 3550 were shown to be related to the genetic male sterile genes. OPA-13 2300 was the characteristic band in B.napus.
