摘要
目的探讨还原型谷胱甘肽(GSH)对脐带间质干细胞生物学特性的影响。方法用MTT法检测脐带间质干细胞活力、细胞计数法绘制细胞生长曲线、流式细胞仪检测细胞周期和表面标志,成骨诱导后组织化学检测碱性磷酸酶(ALP)表达。结果 GSH浓度低于0.15 mg/ml时可促进脐带间质干细胞的增殖,高于此浓度时对细胞的抑制作用随浓度加大而逐步增强。GSH可缩短脐带间质干细胞到达生长平台期的时间并延长细胞传代的代数,细胞周期检测显示S期显著增加,同时GSH对脐带间质干细胞表面标志无明显影响,成骨诱导分化后ALP表达阳性率无明显改变。结论低浓度GSH可促进脐带间质干细胞的增殖,0.15 mg/ml浓度可以达到最佳促进增殖效果,同时对细胞表面标志与向成骨细胞分化的能力无明显影响,可以用于体外对脐带间质干细胞的扩增。
Objective To explore the effects of glutathione on the biological characteristics of human umbilical cord mesenchymal stem cells (hUC-MSCs). Methods The effects of glutathione on hUC-MSC were investigated by the determination of cell activity with MTT assay. Cell suflhce markers and cell cycle were detected by FCM. Differentiation potency of MSC though bone induction and the expression of ALP were examined. Results 0. 15 mg/ml GSH could promote the growth of MSC and raise the proportion of "S" stage without affecting its own characteristics. Conclusions GSH can promote the growth of MSCs without affecting its own characteristics, which suggests that GSH can be use to improve the rate of MSC expansion in vitro.
