摘要
目的:研究热休克处理小鼠肥大细胞瘤细胞P815对细胞内Qa-1表达影响。方法:将体外培养的小鼠肥大细胞瘤细胞P815分为对照组及热休克组(3个温度),共4组:对照组细胞置于37℃的水浴箱中1 h,而后将细胞正常培养4 h;热休克组细胞分别置于41℃、42℃和43℃的水浴箱中1 h,然后恢复至37℃。RT-PCR及流式细胞技术检测分析细胞内Qa-1 mRNA及蛋白水平。结果:热休克可引起小鼠肥大细胞瘤细胞P815胞内Qa-1的mRNA和蛋白水平明显升高,与对照组比较,差异均有统计学意义(均P<0.05),其中43℃热休克处理对Qa-1表达的诱导作用最强。结论:热休克可引起小鼠肥大细胞瘤细胞P815 Qa-1表达上调。
Objective:To determine the effects of heat shock on Qa-I expression in mastocytoma ceils P815 in mice. Methods: The in vitro mice P815 mastocytoma cells were randomly allocated to be treated with 37 ℃ culture for lh followed by conventional cell culture for 4h (control group) and 41 ℃, 42 ℃ and 43 ℃ water bath for lh followed by incubation at 37 ℃ (heat shock groups l, 2 and 3). Reverse transcriptase polymerase chain reaction and flow cytometry were employed to examine the expression Qa-1 mRNA and protein. Results: Heat shock, particularly the incubation at 43 ℃ , yielded significantly increased intracellular expression of Qa-1 mRNA and protein in mastoeytoma cells P815 (both P 〈 0.05) compared with control group. Conclusion: Heat shuck may lead to up-regulation of Qa-1 expression in mastocytoma cells P815.
