摘要
目的:观察红杉醇对2型糖尿病大鼠脂代谢的影响。方法:采用高糖高脂饮食加腹腔注射小剂量链脲佐菌素(Streptozotocin,STZ)诱导2型糖尿病大鼠模型35只,灌服红杉醇(50、25及12.5mg.kg-1.d-1)6周,每天1次。全自动生化分析仪测定血清TG、TC、低密度脂蛋白胆固醇(Low density lipoprotein cholesterol,LDL-C)含量;ELISA法检测血浆游离脂肪酸(Free fattyacids,FFA)、肝脏载脂蛋白A1(ApolipoproteinA1,Apo-A1)和载脂蛋白B1(Apolipoprotein B1,Apo-B1)含量;放射免疫法测定肝组织瘦素(Lep-tin)含量;MASSON染色观察肝脏病理改变。结果:红杉醇能显著降低模型大鼠血脂、FFA含量,下调肝组织Apo-B1及Leptin水平,上调肝组织Apo-A1的含量,减轻肝细胞脂肪变性,减少肝细胞间质和血管旁胶原沉积。结论:红杉醇具有调节2型糖尿病大鼠脂代谢、减轻脂肪肝和肝脏胶原沉积的作用。
ABSTRACT AIM: To observe the effects of sequoyitol on lipid metabolism in rats with type 2 diabetes. METHODS:Type 2 diabetic rats were induced by fed with a high fat and high sugar diet as well as injected with a low dosage of streptozotocin (35 mg·kg^-1). Sequoyitol (12.5, 25, 50 mg·kg^-1·d^-1) was orally administered once a day in 35 model rats for 6 weeks. The contents of fasting triglyceride(TG), total cholesterol(TC)),and low density lipoprotein (LDL) were measured using fully automatic biochemical analyzer. ELISA was used to measure the contents of free fatty acids (FFA) in plasma,and the levels of apolipoprotein A1 (Apo-A1) and apolipoprotein B1(Apo-B1) in liver tissues. The content of leptin was measured with radioimmunoassay. In addition,pathological changes in liver was observed with MASSON staining. RESULTS:Sequoyitol reduced the contents of blood lipids,FFA,Apo-B1 and leptin,elevated the content of Apo-A1. At the same time,sequoyitol ameliorated hepatocellular fatty degeneration, reduced collagen deposition at liver cell interstitial and next to the blood vessels. CONCLUSION:The results show that sequoyitol regulates lipids metabolism and inhibites hepatic steatosis and collagen deposition in type 2 diabetes rats.
