摘要 目的:探讨转酮醇酶样基因1(Transketo-lase-like gene1,TKTL1)在胶质瘤中的表达及意义。方法:收集54例手术切除的胶质瘤标本,6例正常脑组织,通过Realtime-PCR、Western blot及免疫组化方法,从mRNA和蛋白水平检测TKTL1在54例胶质瘤、6例正常垂体组织的表达。结果:Realtime-PCR、Western blot及免疫组化发现TKTL1在胶质瘤组织其表达显著高于正常脑组织(P<0.01),在高级别胶质瘤中和低级别胶质瘤中的蛋白及mRNA表达量两者之间差异具有统计学意义(P<0.01)。结论:TKTL1的表达与肿瘤恶性程度密切相关,表明TKTL1的表达与肿瘤的发生、发展密切相关,提示TK-TL1有可能是预测胶质瘤预后的重要指标。 Objective:To investigate the expression and siguificance of TKTL1 in glioma.Methods:Fity-four gliomas were collected immediately after surgery.Realtime-PCR、Western blot and Immunohistochemistry analysis were performed to determine the expression of TKTL1 in Fity-four gliomas and six normal brain tissues.Results:The expression of TKTL1 in all gliomas was observed to be stronger than that in normal brian by Westerblot and realtime PCR analysis(P〈0.01),IHC showed the expression of TKTL1 was in cytoplasm of glioma,in addition,The expression of TKTL1 in high grade and low grade was different which was significantly statistical (P〈0.01).Conclusion:The results reveal that what were relation of occurring and development contributing to the invasion of glioma,it may be useful to forecast development of glioma by examination.
