不同稀释度A型肉毒毒素局部注射对小鼠神经肌肉的影响 被引量:1

摘要 目的通过观察不同稀释度A型肉毒毒素(botulinum toxin type A,BTX-A)局部注射对小鼠神经肌肉的影响,明确BTX-A局部应用合适的稀释度。方法将25只小鼠随机分为5组,其中4组分别用不同稀释度BTX-A(1.25U/0.1mL、2.50U/0.1mL、5.00U/0.1mL、10.00U/0.1mL)在右侧腓肠肌局部注射,其余1组切断右侧坐骨神经作为阳性对照。左侧均注射生理盐水作为阴性对照。对小鼠进行趾运动评分,检测腓肠肌动作电位后,取腓肠肌称重并进行冰冻切片,行HE染色及免疫荧光检测神经细胞粘附分子(neural cell adhesion molecule,NCAM),电镜下观察腓肠肌肌肉及神经超微结构。结果2.50U/0.1mL及5.00U/0.1mL BTX-A处理组小鼠趾运动评分下降最快、恢复最慢;腓肠肌动作电位阈强度电位增加、最大刺激强度增大、潜伏期延长、动作电位峰值减小,与其他组差异有统计学意义(P<0.001);形态学检测示此2处理组较其他组肌细胞间隙增宽、N-CAM表达增强、肌肉及神经变性明显。结论 2.50U/0.1mL和5.00U/0.1mL BTX-A局部注射起效快、维持作用时间长且去神经支配作用强,它们可能是BTX-A局部注射合适的稀释度,这为BTX-A的临床合理应用提供实验依据。
