金刚健骨片对去卵巢模型鼠TGF-β1及IGF-1表达的影响 被引量:1

The Influence of Jingang Jiangu Tablet on Expression of TGF-β1 and IGF-1 in Ovariectomized Rats

摘要 目的:观察金刚健骨片对去卵巢大鼠骨组织中TGF-β1及IGF-1表达的影响,进一步探讨金刚健骨片治疗绝经后骨质疏松症的作用机制。方法:将健康清洁级雌性SD大鼠40只随机分为5组,即假手术(A)组、金刚健骨片大剂量(B)组、金刚健骨片小剂量(C)组、雌激素(D)组、模型(E)组。所有大鼠常规饲料喂养,造模成功后使用对应药物灌胃治疗12周。治疗结束后处死大鼠采集第三腰椎为标本,采用免疫组化法检测TGF-β1及IGF-1在骨组织中的表达。数据使用SPSS16.0软件统计,检验水准α=0.05.结果:①TGF-β1在骨组织中的表达,B组、D组与E组比较差异有统计学意义(P<0.05);B组与D组比较其差异无统计学意义(P>0.05)。C组与E组比较其差异无统计学意义(P>0.05);C组、E组与A组比较其差异均有统计学意义(P<0.01)。②IGF-1各组间比较差异无统计学意义。结论:①金刚健骨片能使TGF-β1在骨组织中的表达增强,其治疗作用与雌激素治疗相仿,并能修复或者改善骨组织微细结构。②骨组织中IGF-1含量与雌激素关联不明显,金刚健骨片可略提高IGF-1的表达,但无统计学意义。 Objective:To observe the regulating effect of Jingang Jiangu Tablet (JJT) on expression of IGF-1 and TGF-β1 in bone tissue from ovariectomized rats, which might explain the mechanism of JJT in treating postmenopausal osteoporosis. Melhods:Forty female SD rats were randomly divided into 5 groups, namely sham operation group (A), high dose group (B), low dose group (C), estrogen group (D), and model group (E). After treatment, rats were sacrificed. The third lumbar vertebra was collected for detection of TGF-β1 and IGF-1. Results:There was significant difference in TGF {]1 between group B, D and group E (P〈0.05). No difference was found either between group B and group D (P~〉0.05), or between group C and group E (P〉 0.05). Compared with group A, there was significant difference in group C and group E (P〈0.01). There was no remarkable difference in IGF 1 among all groups. Conclusion:JJT can increase TGF-β1 expression in bone tissue, and can repair or improve tissue microstructure. Its efficacy is similar to that of estrogen. There is no obvious association between IGF-1 level and estrogen. JJT can slightly increase IGF1 expression, but the change is insignificant.
