通脑溶栓胶囊对缺血性脑损伤的保护作用 被引量:3

摘要 目的研究通脑溶栓胶囊对缺血性脑损伤的保护作用。方法 SD大鼠按体重随机分为6组:假手术对照组、模型对照组、阳性对照组及通脑溶栓胶囊低、中、高剂量组,前两组灌胃5g/L的CMC-Na,其余各组灌胃等体积对应的受试品混悬液,连续给药30d,末次给药后制备脑缺血模型,观察该药对脑缺血大鼠神经症状、脑组织含水量及脑梗死率的影响。小鼠分为5组:溶媒对照组、阳性对照组及通脑溶栓胶囊低、中、高剂量组,每组10只,雌雄各半。溶媒对照组灌胃5g/L的CMC-Na,其余各组灌胃等体积对应的受试品混悬液,连续给药30d,末次给药30min后断头,记录小鼠喘气时间。结果与模型对照组比较,通脑溶栓胶囊中、高剂量组能明显改善神经症状评分、脑水肿及脑梗死范围,并有统计学意义(P<0.05);低剂量组在改善脑缺血指标上有一定的药理学趋势,但无统计学意义(P>0.05)。与溶媒对照组比较,通脑溶栓胶囊低、中、高剂量组可延长小鼠断头后喘气时间,并有统计学意义(P<0.05或P<0.01)。结论通脑溶栓胶囊能保护缺氧对脑组织的损伤,提高脑组织耐缺氧的能力。 Objective To study the protective effect of Tong-nao thrombolysis capsule on ischemic brain injury.Methods SD rats were randomly divided into six groups according to weight: sham group,model group,positive control group,and Tong-nao thrombolysis capsule low,medium and high dose groups.The first two groups were intragastrically administered 5 g/L CMC-Na while the other groups were intragastrically administered the corresponding volume of suspension for 30 d.The cerebral ischemia models were made after the final drug administration to observe the influences of the drug on nerve symptom,water content in brain tissues and the rate of cerebral infarction in cerebral ischemia mice.The mice were divided into 5 groups: solvent control group,positive control group,and Tong-nao thrombolysis capsule low,medium and high dose groups(10 in each group,males and females in half).The solvent control group was intragastrically administered 5 g/L CMC-Na;the other groups were intragastrically administered the corresponding volume suspension continuously for 30 d.The mice were beheaded 30 min after the final drug administration to record gasping time.Results Compared to the model group,Tong-nao thrombolysis capsule medium and high dose groups differed significantly in neurologic symptoms scores,cerebral edema and brain infarction range(P0.05).Low dose group had a certain pharmacology trend in improving cerebral ischemia index,but without statistical difference(P0.05).Compared with the solvent control group,Tong-nao thrombolysis capsule low,medium and high dose groups had significantly prolonged gasping time after beheaded(P0.05 or P0.01).Conclusion Tong-nao thrombolysis capsule can protect the brain fromhypoxia-induced damage and improve anoxia tolerance capability of brain tissues.
