摘要
目的利用小干扰RNA(siRNA)抑制肝癌细胞雌激素受体(ER)基因的表达,观察其受抑制对细胞增殖、凋亡及侵袭能力的影响。方法通过构建ER—siRNA慢病毒载体系统转染人肝癌细胞Hep3B细胞,将实验分为3组:空白对照组、阴性对照组及siRNA组,应用Westernblot检测ER基因沉默的效果,用噻唑蓝(MTr)方法检测细胞增殖,用流式细胞仪观察肿瘤细胞凋亡,用Transwell侵袭实验检测检测Hep3B细胞体外侵袭力的改变。结果成功构建ER基因的慢病毒siRNA载体,感染肝癌细胞明显抑制ER蛋白表达,抑制率56.64%;空白对照组、阴性对照组及小干扰RNA组MTT的吸光度(A)值分别0.68±0.11—060±0.13和0.49±0.10(P〈0.05);流式细胞检测s期的细胞比例分别为(35.23±4.68)%、(30.95±3.17)%和(21.88±2.13)%(P〈0.05);迁移实验细胞数分别为(97.66±5.03)、(90.67±3.79)和(37.00±10.14)个(P〈0.05)。结论重组ER—siRNA慢病毒可抑制Hep3B细胞ER蛋白表达水平,抑制细胞增殖,促进细胞凋亡,降低肿瘤细胞侵袭能力。
Objective To observe the effect on the growth and invasive capacity of hepatocarcino- ma cells by inhibiting the expression of estrogen receptor (ER) gene. Methods ER-small interfering RNA (siRNA) lentivirus vector was constructed and transfected into Hep3B cells. Three groups were set up: control group (CON), negtive control group (LV) and ER-siRNA lentivirus group (ER-siRNA). Westem blotting was used to detect the expression of ER protein. The proliferation of Hep3B cells was measured by using methyl thiazol tetrazolium (MTT) assay. Apoptosis was observed by using flow cytometry. The inva- sive activity of infected Hep3B ceils was analyzed by using Transwell invasion assay. Results Recombinant lentiviral vector expressing siRNA targeting ER gene was successfully established. Western blotting showed that the ER protein expression in Hep3B cells infected with ER-siRNA lentivirus was decreased by 56. 64%. In CON group, LV group and ER-siRNA lentivirus group, the A values were O. 68 _±0. 11, O. 60 s0. 13 and 0. 49 sO. 10 respectively (P 〈0. 05) ; the S-phase cell fraction by flow cytometry was (35.23 ±4. 68)%, ( 30. 95 ±3. 17 ) % and ( 21.88 ± 2. 13 ) % ( P 〈 O. 05 ) ; and the number of Hep3 B cells in invasion assay were 97.66 ± 5.03, 90. 67 ± 3.79 and 37.00 ±10. 14 (P 〈 0. 05 ), respectively. Conclusion ER-siRNA lentivirus could inhibit the expression level of ER protein, induce apoptosis, reduce proliferartion and inva- sion capacity of Hep3B ceils.
