Ⅰ~Ⅲ期甲状腺乳头状癌中微小RNA的差异表达及其功能 被引量:8

Differential expression profiling and functional analysis of microRNAs in stage I-III papillary thy- roid carcinoma

摘要 目的探讨Ⅰ~Ⅲ期甲状腺乳头状癌组织和癌旁组织微小RNA(miRNAs)分子表达谱的差异。方法收集Ⅰ、Ⅱ、Ⅲ期甲状腺乳头状癌及相应癌旁组织各1例,利用miRNA芯片技术,分别检测3例甲状腺乳头状癌与癌旁组织中miRNAs的表达,对按照临床分期呈顺序上调或下调的miRNAs进行分析;采用实时定量聚合酶链反应(Real—timePCR)对部分miRNAs芯片结果进行验证;通过靶基因预测软件预测部分miRNAs可能调控的靶基因,并分析其生物学功能。结果与癌旁组织比较,在Ⅰ~Ⅲ期标本中均呈现显著性差异表达的miRNA分子20个;按照临床病理分期,其中在甲状腺乳头状癌中呈现顺序上调的miRNA分子有8个,呈现顺序下调的有5个。结论Ⅰ-Ⅲ期甲状腺乳头状癌和癌旁组织之间存在差异表达miRNA分子,它们的异常表达可能在甲状腺乳头状癌发生发展过程起重要调控作用。 Objective To explore the microRNA differential expression profile between I-]lI papillary thyroid carcinoma ( PTC ) and peritumoral tissues so as to provide rationals for the role of microRNA in the pathogenesis of PTC. Methods Samples of stage Ⅰ~Ⅲ and m PTC and peritumoral tissues ( n = 1 for each) were collected. The expression levels of microRNAs were examined by using mieroRNA microarray chip. MicroRNAs that were sequentially upregulated or downregulated with clinical staging were further analyzed. Some of miRNAs microarray results were verified by real-time quantitative polymerase chain reac- tion (real-time PCR). Some potential miRNAs regulating target genes were forecasted by using function prediction software, including their preliminary biological functions. Results Compared with peritumoral tissues, 20 microRNAs exhibited significant differential expression in PTC. In addition, 8 microRNAs were up-regulated sequentially in stage Ⅰ~Ⅲ PTC tissues, and 5 mieroRNAs were down-regulated sequentially. Conclusion There is a differential expression of microRNA between stageⅠ~Ⅲ PTC and peritumoral tissues, which may be related with the occurrence and metastasis of PTC.
