摘要 目的:在人的非小细胞肺癌细胞A549中通过降低K-Ras表达研究其在自噬中的作用。方法:磷酸钙方法转染K-Ras-shRNA,Western blot检测自噬标志物的变化。结果:在A549细胞中,通过K-Ras-shRNA慢病毒转染显著地降低了K-Ras的蛋白水平,同时自噬标志物LC3II水平降低,自噬调控分子Beclin-1水平降低。结论:K-Ras knockdown引起细胞自噬水平的降低。 Objective: To explore the contribution of K-Ras to autophagy after K-Ras knockdown. Methods: Calcium phosphate method and Western blot were used to transfect K-Ras-shRNA and to detect the changes of autophagy marker, respectively. Results: In A549 cells, protein level of K-Ras was reduced significantly by K-Ras-shRNA and autophagy marker LC3II decreased after K-Ras knockdown, so did autophagy regulating molecule Beclin-1. Conclusion: K-ras knockdown can result in the reduction of autophagy level.
