摘要
目的研究二甲双胍对高脂高糖饮食联合小剂量链脲佐菌素(STZ)诱导的糖尿病大鼠肝脏脂肪变性的影响。方法采用高脂高糖饲料喂养雄性SD大鼠6周后,一次腹腔注射STZ 40 mg/kg诱导糖尿病大鼠模型。实验分为正常对照组、模型组、二甲双胍组。二甲双胍灌胃给药4周后观察其对糖尿病大鼠空腹血糖(FBG)、空腹胰岛素(FINS)、胰岛素抵抗指数(HOMA-IR)、游离脂肪酸(FFA)、甘油三酯(TG)、总胆固醇(CHOL)水平的影响,进行肝脏组织HE染色,观察其对糖尿病大鼠肝脏脂肪变性的影响。结果与正常对照组比较,模型组FBG、FINS、HOMA-IR、FFA、TG明显升高(P<0.05),肝脏组织发生脂肪变性;与模型组比较,二甲双胍组FBG、FINS、HOMA-IR显著降低(P<0.05);二甲双胍能减轻糖尿病大鼠肝脏脂肪变性。结论二甲双胍能减轻糖尿病大鼠肝脏脂肪变性,其机制与改善胰岛素抵抗有关。
Objective To investigate the effects of metformin on liver fatty degeneration in diabetic rats. Methods Male Spragye - Dawley rat model of diabetes was established with high - fat diet for 6 weeks followed by intraperitoneal injection of Streptozotocin (40 mg/kg). Rats were randomly divided into nor- mal group, model group and metformin group. After intragastric administration of metformin for 4 weeks, the fasting plasma glucose (FBG) ,fasting insulin (FINS), free fatty acids (FFA), triglyeride (TG) and cholesterol (CHOL) were determined and homeostasis model assessment of insulin resistance (HOMA- IR) were calculated. Rat liver fatty degeneration was observed after Hematein Eosin stai- ning. Results Compared with the normal group, the levels of FBG, FINS, FFA, TG and HOMA - IR were significantly increased and liver fatty degeneration was observed in model group. Compared with the model group, metformin significantly attenuated rat liver fatty degeneration and the elevations of FBG, FINS and HOMA -IR. Conclusion Metformin could reduce the liver fatty degeneration through the improvement of insulin resistance.
