保胎液对胚泡着床障碍小鼠着床位点数及分布形态影响的实验研究 被引量:1

Experimental Research of the Impacts of Baotaiye on the Implantation Rate and Distribution Morphology for the Mice with Blastocyst Implantation Disorder

摘要 目的观察保胎液对胚泡着床障碍模型小鼠着床位点数及分布形态的影响。方法采用米非司酮小鼠胚泡着床障碍模型。将孕鼠随机分为正常组、模型组、西药对照组和中药(高、中、低三种剂量)治疗组,从妊娠第1天起分别给予生理盐水、地屈孕酮和保胎液(高、中、低三种剂量)灌胃治疗。于妊娠第4天上午皮下注射米非司酮,每只0.08 mg。正常组不予任何处理。于妊娠第5天腹腔静脉注射1%浓度的Trypan Blue 0.3 mL,观察10 min,取出子宫,观察蓝染点,计算胚泡着床位点数。结果中药高、中、低治疗组,西药对照组,模型组小鼠胚泡着床位点数均低于正常组(P<0.05);与模型组比较,中药各治疗组、西药组小鼠着床位点数明显提高(P<0.05)。结论保胎液能够增加着床障碍小鼠的胚泡着床位点数,从而有利于胚泡的着床,提高妊娠率。 Objective To observe the impacts of baotaiye on the implantation rate and distribution morphology for the model mice with blastocyst implantation disorder. Methods Mifepristone was used to prepare the model of blastocyst implantation disorder. The pregnant mice were randomized into a normal group, a model group, a western medicine control group and Chinese medicine (high -dose, middle -dose and low - dose ) treatment groups. Since the i st day of pregnancy, the physiological saline, dydrogesterone and baotaiye ( high - dose, middle - dose and low - dose) were used for lavage treatment separately. Since the morning on the 4th day ,the subcutaneous injection of mifepristone was done ,0.08 mg for each mouse. No any treatment was given for the mice in the normal group. On the 5th day of pregnancy, the abdominal intravenous injection of 1% trypan blue 0.3 mL was done. After observed for 10 rain, the uterus was collected. The blue - stained spots were observed and the blastocyst implantation rate was calculated. Results The blastocyst implantation rate for the mice in the high - rate, middle - rate and low - rate Chinese medicine treatment groups, western medicine group and model group was all lower than that in the normal group separately (P 〈 0.05 ). Compared with the model group separately, the blastocyst implantation rate in each Chinese medicine treatment group and western medicine group was increased apparently( P 〈 0.05 ). Conclusion Baotaiye in- creases the blastoeyst implantation rate for the mice with implantation disorder so that it contributes to the blastocyst implantation and improves the pregnancy rate.
