氧化应激心肌细胞prohibitin表达与分布变化及其生物学意义 被引量:1

摘要 目的分析应激心肌细胞prohibitin被氧化过程中的表达与分布变化,及其生物学意义。方法用过氧化氢对进行体外培养的大鼠心肌细胞干扰,将其作为应急心肌细胞氧化的模型;并用科学方法检查细胞培养溶液中乳酸脱氢酶的活跃性以及运用MTT实验来证实大鼠心肌细胞的受损情况;并利用蛋白质印迹技术来检查心肌细胞prohibitin在被氧化应激时,蛋白质的表达和分布变化情况,通过ATPase实验类测定线粒体的活性从而推断出线粒体氧化磷酸化的作用;最后运用流式细胞技术来判定线粒体的跨膜电势。结果氧化后乳酸脱氢酶的活跃性明显高于对照组,但研究组细胞的成活几率远远小于对照组;通过ATPase实验类测定线粒体的活性下降了大约60个百分点;线粒体prohibitin的表达情况优于对照组。结论应激心肌细胞prohibitin被氧化后表达水平生物体的自我调节能力加强,并且开始慢慢向线粒体移动,氧化应激使心肌细胞线粒体受损,从而功能出现问题。
