摘要
采用荧光定量RT-PCR和间接免疫荧光方法对泌乳期高乳蛋白率、泌乳期低乳蛋白率以及干奶期的荷斯坦奶牛乳腺中LAT1和4F2hc的表达变化与定位变化进行研究,结果表明,泌乳期高乳蛋白率的荷斯坦奶牛乳腺中LAT1特异在乳腺腺泡上皮细胞基底侧表达,4F2hc在乳腺腺泡上皮细胞中表达,LAT1和4F2hc的表达量均显著高于泌乳期低乳蛋白率的荷斯坦奶牛和干奶期的荷斯坦奶牛,预示在奶牛乳腺泌乳过程中,LAT1和4F2hc参与调控乳蛋白的生物合成速率。
In this research, the expression and localization of LAT1 and 4F2hc in lactation dairy cow mammary gland and in dry period mam- mary gland were detected by qP, T-PCR and immunofluorescence. The results showed that in lactation mammary gland with high protein, LAT1 was located in the basement membrane of mammary epithelial cells. 4F2hc was expressed in the cytoplasm of mammary epithelial cells. The expression of LAT1 and 4F2hc in lactation mammary gland with high protein were higher than those in lactation mammary gland with low protein and dry period mammary gland. It indicated that LATland 4F2hc may play an important role to regulate milk protein synthesis.
