摘要
目的观察通心络超微粉对脑缺血大鼠微血管及血流灌注量的影响。方法将48只健康SPF级SD大鼠随机分为假手术组、模型组、通心络大剂量组[1.6g/(kg·d)]、通心络中剂量组[0.8g/(kg·d)]、通心络小剂量组[0.4g/(kg·d)]、尼莫地平组[10mg/(kg·d)],每组8只。采用开颅结扎法阻断大鼠一侧大脑中动脉制备局灶性脑缺血大鼠模型,假手术组大鼠只挑开脑膜,不结扎血管。各用药组均在动物苏醒后灌胃,假手术组和模型组给予等体积生理盐水。给药14天后,利用激光扫描共聚焦显微镜观察各组大鼠脑组织微血管形态、直径及血流灌注量。结果与模型组比较,各用药组大鼠大脑皮层微血管均呈现迂曲不规则形状,其中通心络大剂量组血管明显呈迂曲、不规则,微血管分支数目增多,血管之间靠拢形成连接。模型组缺血侧40%微血管直径在5~6μm之间;通心络大剂量组缺血侧41%微血管直径在2~3μm之间;通心络中剂量组缺血侧40%微血管直径在3~4μm之间;通心络小剂量组缺血侧40%微血管直径在4~5μm之间。与模型组比较,通心络大、中剂量组、尼莫地平组显著促进缺血侧脑组织皮层区微血管血流灌注量增加(P<0.01)。结论通心络超微粉可促进脑缺血大鼠脑缺血区微血管新生,增加缺血脑组织血流灌注量。
Objective To observe the effect of Tongxinluo Supertine Powder (TSP, superhne powder to free the laeart vessels) on microvessels and Blood perfusion in cerebral ischemia rats. Methods Totally 48 healthy SPF SD rats were randomized into sham operation group, model group, TSP high-dose group (1.6g/kg . d), TSP middle-dose group (0.8g/ kg . d), TSP low-dose group (0.4g/kg . d) and nimodipine group (10mg/kg . d), with 8 in each. The focal cerebral ischemia models were established with craniotomy ligation of one side of the middle cerebral artery. The rat meninges in the sham operation group were picked open without vascular ligation. Each treatment group was given intragastric administration after waking and the sham operation group and model group were given the same volume of normal saline. The morphology, diameters and blood perfusion of cerebral microvessels were observed with laser scanning confocal microscopy on the 14th day of administration. Results Comparing with the model group, the cerebral cortex microvessels in each treatment group showed a tortuous and irregular shape and the cerebral cortex microvessels in the TSP high dose group showed obviously irregular shape, numerous microvascular branch and close connection between microvessels. The diameter of 40 % microvascular of ischemic side of the rats in the model group was 5 to 6 tim. The diameter of 41% microvaseular of ischemic side of the rats in the TSP highdose group was 2 to 3μm. The diameter of 40% microvascular of ischemic side of the rats in the TSP middle-dose group was 3 to 4μm. The diameter of 40% microvascular of ischemic side of the rats in the TSP low-dose group was 4 to 5 μm. Comparing with the model group, the blood perfusion in the TSP high-dose group, TSP middle-dose group and nimodipine group was significantly increased (P〈 0.01 or P〈 0.05). Conclusion TSP can promote angiogenesis in cerebral cortex after focal cerebral ischemia and increase the blood perfusion.
