miR-335表达失调对前列腺癌细胞株LNCaP生物学的影响 被引量:5

摘要 目的:探讨miR-335表达失调对前列腺癌细胞株LNCaP的生物学影响。方法:通过载体构建、细胞转染及筛选、荧光定量PCR验证等实验方法构建miR-335过表达前列腺癌细胞株LNCaP,荧光定量PCR验证,CCK8法检测miR-335过表达细胞株及miR-335阴性表达细胞株的生长趋势。结果:成功构建miR-335过表达前列腺癌细胞株LNCaP,miR-335在过表达细胞株中的表达量显著低于阴性表达细胞株(P=0.001),同时过表达细胞株的生长速度也显著慢于阴性表达细胞株(P<0.01)。结论:miR-335过表达可抑制前列腺癌细胞的体外增殖,在前列腺癌的发病机制中扮演候选抑癌基因的角色。
