白藜芦醇对小鼠神经元线粒体mtDNA拷贝数及相关基因表达的影响

Effects of resveratrol on the level of mtDNA and the expressions of mitochondrial corresponding genes in mitochondria in cerebral cortex neurons of mice

摘要 目的:研究白藜芦醇(Res)对体外培养的小鼠大脑皮层神经元线粒体形成的作用。方法:培养神经元分为3组:空白对照组、白藜芦醇组(Res组)、二甲基亚砜组(DMSO组)。处理7d后检测培养神经元ATP含量和线粒体DNA拷贝数,采用Real timePCR方法检测线粒体形成相关分子Tfam、NRF-1、NRF-2和PGC-1α的表达情况。结果:Res处理能够明显增加体外培养小鼠神经元ATP含量,Res组ATP水平明显高于DMSO组(P<0.01);Res组mtDNA拷贝数明显增加(P<0.001),而DMSO组与空白对照组组间mtDNA拷贝数无明显差异。此外,Res明显上调了线粒体形成相关分子Tfam、NRF-1、NRF-2和PGC-1α的基因表达(P<0.001)。结论:Res处理可能具有通过影响mtDNA拷贝数及线粒体形成相关基因的表达,促进小鼠神经元线粒体形成的潜能。 Objective To investigate the effects of resveratrol on the biogenesis of mitochondria in cerebral cortex neurons of mice cultured in vitro. Methods The neurons cultured in vitro were divided randomly into sham group, dimethyl sulfoxide (DMSO) group and resveratrol group. After 7 days of cuhivation, the ATP content and mtDNA level in neurons were measured, and the expressions of Tram, NRF-1, NRF-2 and PGC-1α were evaluated by real time PCR. Results As compared with those in DMSO group, the ATP content and mtDNA level in neurons in resveratrol group were significantly increased (P 〈 0.01 and P 〈 0.001). In addition, the mRNA expressions of Tram, NRF-1, NRF-2 and PGC-1α were enhanced by resveratrol (P〈 0.001). Conclusion Resveratrol might potentially promote the biogenesis of mitochondria in cerebral cortex neurons of mice cultured in vitro through affecting the level of mtDNA and the expressions of mitochondrial corresponding genes.
