糖尿病大鼠缺血再灌注脑组织中FADD、Daxx的表达及其意义 被引量:2

摘要 目的:研究Fas相关死亡域蛋白(FADD)、死亡域相关蛋白(Daxx)在糖尿病缺血再灌注脑组织损伤中的作用。方法:36只健康雄性SD大鼠按随机数字表法分为3组:假手术组、脑缺血再灌注组和糖尿病脑缺血再灌注组。大鼠腹腔内注射链脲佐菌素制作糖尿病模型,之后再应用栓线法建立大鼠局灶性脑缺血再灌注模型。于缺血1h再灌注12h后对各组大鼠进行神经功能缺损评分,TTC染色测定梗死体积,Tunel法检测细胞凋亡数目,免疫组化染色及Western blot检测缺血半暗带侧皮质区FADD、Daxx的表达。结果:与脑缺血再灌注组相比,糖尿病脑缺血再灌注组的神经功能缺损评分、梗死面积及凋亡细胞数均有明显增加,差异均有统计学意义(P<0.05)。糖尿病脑缺血再灌注组免疫组化染色检测FADD灰度值为110.18±4.21、Daxx为101.26±3.27;Western blot检测FADD蛋白定量为1.191±0.041、Daxx为1.389±0.052。脑缺血再灌注组免疫组化染色检测FADD灰度值为131.46±3.24、Daxx为123.16±2.19;Western blot检测FADD蛋白定量为1.035±0.032、Daxx为1.146±0.083。与脑缺血再灌注组相比,糖尿病脑缺血再灌注组FADD及Daxx蛋白的表达均明显增高,差异有统计学意义(P<0.05)。结论:FADD、Daxx可能参与了糖尿病加重脑缺血再灌注损伤的病理生理过程,FADD、Daxx表达增加可能是糖尿病加重脑缺血再灌注损伤的机制之一。
