摘要 目的为建立栀子苷的免疫学分析方法,首先合成栀子苷的人工完全抗原。方法采用高碘酸钠氧化法分别合成栀子苷免疫抗原(GEN-BSA)和包被抗原(GEN-PLL)。结果优化了栀子苷抗原的合成条件,经紫外光谱检测栀子苷成功与BSA/PLL偶联,偶联比可达到11∶1,免疫小鼠后血清抗体效价达到1∶12 800。结论成功合成栀子苷完全抗原,可用于下一步单克隆抗体的制备。 Objective To establish a immunoassay for analyzing geniposide (GEN), and synthesize the artificial complete antigen of GEN. Methods The immunizing antigen (GEN-BSA) and coating antigen (GEN-PLL) were synthesized respectively by applying sodium periodate oxidation method. Results The synthesis conditions of GEN antigen was optimized. After the detection of ultraviolet spectrum, GEN was successfully conjugated with BSA/PLL and the conjugation ratio was 11: 1. The serum antigen titer had reached to 1:12 800 in immunized mice. Conclusion The complete antigen of GEN has been successfully synthesized, which can be used further for preparing monoclonal antibody.
