VEGF、p53在精原细胞瘤中的表达及相关性分析

Relationship between expression of VEGF and p53 genes and clinicopathologicalparameters in seminoma of testis

目的通过对人血管内皮生长因子(vascular endothelial cell growth factor,VEGF)、p53基因的研究,从分子水平上研究精原细胞瘤恶变过程中癌基因、抑癌基因表达的改变,为早期明确诊断和治疗精原细胞瘤提供分子生物学上的依据。方法应用免疫组化辣根过氧化物酶标记的链霉卵白素(streptavidin-peroxidase,S-P)法,检测VEGF、p53基因在38例精原细胞瘤中的表达。应用PCR-单链构象多态性(PCR-SSCP)法,检测p53在精原细胞瘤中的突变及缺失情况。结果 VEGF在精原细胞瘤临床分期Ⅰ期的阳性表达率为33.33%,Ⅱ期和Ⅲ期的阳性表达率分别为90.91%和100%,p53在精原细胞瘤临床分期Ⅰ期的阳性表达率为93.33%,Ⅱ期与Ⅲ期的表达率分别为54.55%和50%,VEGF和p53基因在精原细胞瘤中的表达无相关性。p53在精原细胞瘤中存在突变,突变率为45.45%。结论 VEGF在精原细胞瘤的发生中发挥着重要作用;p53在精原细胞瘤的发生和发展过程中,存在着基因表达的异常。p53在调控精原细胞瘤发生过程中,主要在临床发生的早期起重要作用;精原细胞瘤中,p53与VEGF不存在相关性。

Objective To investigate the tumor and tumor suppressive genes in the carcinogenesis of seminoma of testis byex- ploring the expression of VEGF and p53 ,and to provide the molecular biologic basis for the early identified diagnostic and treatment of seminoma. Methods 38 paraffinembeded specimens of seminoma were collected. IHC was used to detect the expression of protein of VEGF and p53. PCR-SSCP was used to detect the mutation and deletion of p53. Results The positive expression rates of VEGF in seminoma clinical stage I , II, III were 33.33% ,90.91% , 100% , respectively. The positive expression rate of p53 in seminoma clinical stage I , U, m were 93.33% ,54.55% ,50%. There was no correlation between expression of VEGF and p53 in seminoma. The muta- tion rate in seminoma was 45.45%. Conclusion The VEGF and p53 play important roles in the development and progression of semi- noma. There is an altered expression of p53 in the progression of seminoma and the p53 gene plays important roles in the early stage. No relationship between p53 and VEGF was found.
