PDI单抗对创伤性深静脉血栓形成大鼠体内组织因子活性的影响

PDI Monoclonal Antibody Have Influence on the Activity of Tissue Factor Which Exist in the Rats that Get Traumatic Deep Venous Thrombosis

目的观察PDI(protein disulfide isomerase蛋白质二硫键异构酶)单抗对创伤性深静脉血栓形成大鼠体内组织因子(tissue factorTF)活性的影响,初步探讨PDI蛋白在组织因子活化中的作用。方法选择SD大鼠40只,随机平均分为实验组和对照组。实验组大鼠预先注射PDI单抗,对照组不加干预。钳夹法损伤实验组和对照组大鼠的右股静脉,制造大鼠创伤性深静脉血栓模型,造模后立即抽血,抗体夹心酶联免疫吸附试验(ELISA)检测大鼠体内TF含量,发色底物法检测TF促凝活性,并于第3、10、25h分别抽血,利用ELISA法检测大鼠体内D-二聚体含量,25h后解剖大鼠右股静脉观察血栓形成情况。结果对照组大鼠血清中TF含量及活性均高于实验组(P<0.01),3次抽血实验组大鼠血D-二聚体含量无明显变化,对照组大鼠成增高趋势,解剖后发现实验组大鼠股静脉血栓形成范围、栓子大小及患肢肿胀程度均小于对照组。结论 PDI单抗可以抑制大鼠创伤性深静脉血栓的形成,PDI蛋白在组织因子活化的活化过程起重要作用。

Mothods 40 SD rats were randomly divided into experimental and control groups, the experimental rats pre-injection PDI mAb, control group without intervention. Forceps injury the right femoral vein of rats, both experimental and control groups, manufacturing traumatic deep vein thrombosis model, blood immediately after modeling, antibody sandwich enzyme-linked immunosorbent assay detection rats TF content, chromogenic substrate assay TF procoagulant activity, At 3, 10 and 25hour respeetively for blood, rats measured by ELISA D-dimer. Anatomical rat right femoral vein observed thrombus formation after 120h. Results TF content and activity of the experimental group were higher than in the control group serum（P〈0.01）, D-dimer of experimental rats showed no significant change, The control group of rats into the trend of increasing, anatomical found that the range of experimental rats femoral vein thrombosis, embolus size and degree of limb swelling is less than the control group. Conclusion PDI monoclonal antibody can inhibit the formation of deep vein thrombosis in rats with traumatic,PDI protein plays an important role in the tissue factor activation of the activation process.
