新生期母婴分离致大鼠海马细胞凋亡的信号机制 被引量:6

摘要 目的:研究新生期母婴分离致大鼠海马细胞凋亡的信号机制。方法:将大鼠仔鼠随机分为母婴分离(MS)组(仔鼠从生后第2~21d每天母婴分离3h)和对照(N)组(仔鼠不做任何处理)。将两组仔鼠于分离后第7、14和21d断头取脑,固定、切片,采用Tunel法和免疫组化SP法观察海马神经元凋亡和半胱氨酸蛋白酶-3(Caspase-3),小窝蛋白(Caveo-1)在海马细胞中的表达。结果:与对照组相比,MS组仔鼠在分离后7d、14d和21d神经元细胞凋亡均增多(22.62±0.68)、(12.23±0.97)和(8.14±0.93),(P〈0.01);Caspase-3表达:7d(0.29±0.03)、14d(0.22±0.01)、21d(0.16±0.01)均增多(P〈0.01);小窝蛋白(Caveo-1)在神经胶质细胞中的表达:除7d(0.13±0.01),(P〉0.05)外,14d(0.19±0.01)、21d(0.24±0.01)均减少(P〈0.05)。结论:新生期母婴分离可导致大鼠发育过程中海马神经细胞凋亡和Caspase-3蛋白表达增多,小窝蛋白(Caveo-1)表达减少,这可能与生命早期母婴分离导致成年后的行为发育异常有关。 Objective:To explore the signal mechanism of maternal separation(MS) stimulation in neonatal period on hippocampus apoptosis of developing rats.Methods: Neonatal rats were divided randomly into(MS) group and normal control(N) group.Rats in MS group were separated from the dam for 3h/d(8∶00~11∶00am)on postnatal days 2~21 for 20 consecutive days.Rats in normal control group were not treated and remained in the home cage with the dam.Rats in the two groups were sacrificed to get biains and prepare sections on day 7,14and 21 respectively after separation.Tunel and immuno-histochemistry were used to detect the apoptosis of cells and the protein expressions of Caspase-3 and Caveo-1.Results: Campared with the normal control group,the apoptosis [7d(22.62±0.68),14d(0.22±0.01),21d(0.16±0.01),(P0.01)]in hippocampus were all increased on day 7,14 and 21 resectively after separation;And the expression of Caspase-3[7d(0.29 ±0.03),14d(0.22±0.01),21d(0.16 ±0.01),(P 0.01)]were also increased in above all,but the expressions of Caveolin-1 were decreased on 14d(0.19±0.01),21d(0.24±0.01),(P0.01),except 7d(0.13±0.01)(P0.05).Conclusion: Maternal separation in neonatal period increase the hippocampal neuron apoptosis and the expression of Caspase-3 and decrease the expression of Caveolin-1.Which are possibly related to the mature rats' abnormal behavior in the condiction of consecutive maternal separation in early life.
